Noradrenaline-dopamine disinhibitors are drugs that act by antagonising the serotonin 5-HT2C receptors, inhibiting the noradrenaline and dopamine release in the brain. Agomelatine is the only drug in this class.

Structurally closely related to melatonin, Agomelatine is is comparable to the SSRI antidepressant Paroxetine, a powerful drug, in treatment of Major Depressive Disorder (MDD). Interestingly, the drug exhibits few if any side effects, including the sexual side effects that usually come with many antidepressants. It also improves the quality of sleep that the patient experiences, without presenting daytime drowsiness. In terms of efficiency in combatting Major Depressive Disorder (MMD), Agomelatine has also been shown to be more effective than the SSRI antidepressant Sertraline. On top of this, the drug also has anxiolytic properties. Agomelatine has been shown to have a more favourable withdrawal process than most other antidepressants.
